Acquiring the image

• On the microscope stand - change the light path to

BASEPORT

• In Volocity - select channel from pre-set (1)

• Refocus / re-centre (microscope focus and joystick)

• Then adjust:

2. Exposure time,

3. Binning

4. (and lamp brightness in brightfield channel)

5. NB: Make sure "Auto Contrast" is off!!!

6. Save settings

• Repeat for each required channel

Useful tips:

• Clicking the "Freeze" button in the software will

keep your actual live image frozen on the

screen; the acquisition stops and the shutters

will close, preventing your sample from

bleaching.

• Switch on the contrast range display

(contrast icon on top menu bar).

Left-Click and drag the mouse left

over the image to compress the

displayed range (and right to

expand it). This does not change

the raw data but just the way the

image is displayed on screen

(including the file preview window).
