2.4.5 Reflected light fluorescence
requires the use of the Axiovert 25 CFL microscope stand.
• Select a point on the specimen in transmitted light brightfield or phase contrast. To do this, switch
the reflector mount (2Ć22ā/ā10) to free passage, switch on the halogen lamp (2Ć22ā/ā2) and move the
phase slider (2Ć22ā/ā3) to the middle position (brightfield) or to the position with the ring
diaphragm (phase contrast).
• Switch on the HBO 50 lamp (2Ć22ā/ā5) by means of the power supply unit, but block the light path
using the additional filter slider (2Ć22ā/ā6).
• After selecting the specimen position, cover the transmitted light beam path by means of the
cover plate (2Ć22ā/ā11) in the Phā/ā3Ćfold slider (2Ć22ā/ā3) or switch off the halogen lamp.
• With the reflector mount (2Ć22ā/ā10), slide in the required filter combination (2Ć22ā/ā9) and release
the light path by pulling the additional filter slide (2Ć22ā/ā6).
• By means of the push rod (2Ć22ā/ā7), close the luminous field diaphragm until it is visible in the
image, move it to the middle position by means of the centring screws (2Ć22ā/ā8) and open it up
to the edge of the field of view.
• Additional excitation filters with a diameter of 25 mm, which must be held by means of zero
rings, can be inserted in the additional filter slider (2Ć22ā/ā6) of the reflected light unit Fl.
• The additional filter slider (2Ć22ā/ā6) allows no light to pass in the center stop position.
